Separation of methylated free bile acids from their taurine and methyl glycine conjugates by thin-layer chromatography.

چکیده

Class separation of methylated free bile acids from bile acids conjugated with taurine and methylglycine was accomplished using a solvent system of 2,2,4-trimethylpentane-absolute ethanol 10:1 (v/v). By developing a silica thin-layer plate two times with solvent in a Brinkmann sandwich tank, the difficult resolution between methyl cholate and methyl glycolithocholate was achieved. Evidence is presented that this separation system may be useful as a preparative step in the analysis of bile acids by gas-liquid chromatography or high pressure liquid chromatography.--Bolt, M. J. Alkaline solvent systems for thin-layer chromatography of bile acids.

Thin-layer chromatographic separation of the common bile acids and their taurine and glycine conjugates in chloroform-methanol-ammonia is reported. An alkaline system offers advantages for the separation and nondestructive staining of bile acid conjugates.

Silicic acid thin-layer chromatography of conjugated and free bile acids.

The separation of free bile acids, taurine conjugates, and glycine conjugates from one another was achieved by thin-layer chromatography, using a solvent system of isopropanol glacial acetic acid 93:7 in a solvent system of hexane-methylethylketone-glacial acetic acid 56:36:8 (v/v), it was possible to separate cholic, chenodeoxycholic, deoxycholic, and lithocholic acids. Spectrofluorimetric determination of individual bile acids in biological fluids: duodenal content and bile.

A micromethod is described for the separation and quantitative determination of individual bile acids in duodenal content and bile. After a mild saponification procedure for splitting the taurine and glycine conjugates, the products are separated by thin-layer chromatography on silicagel layer with the solvent system //0-octane-ethyl acetate-acetic acid (60 ml + 35 ml + 5 ml) and determined by ...

A method for the separation of glycine-conjugated bile acids as a group from taurine-conjugated bile acids.

Ileal disease, liver disease, and other pathologic conditions have been shown to affect the glycine:taurine ratio (1). The procedure used for determining the glycine:taurine ratio is to separate the individual conjugated bile acids on thin-layer chromatograms and estimate the quantities of each bile acid to get the ratio (2, 3). Sometimes two or more solvent systems are needed to separate the i...

Determination of individual conjugated bile acids in human bile.

A method has been developed and validated for the determination of the six major conjugated bile acids, cholesterol, and total phospholipids in bile of human subjects previously injected with 4-(14)C-cholesterol. The procedure is designed for use with 5-10 ml of duodenal or T-tube bile and eliminates difficulties associated with existing methods for bile acid determination, in particular the re...
